General description
Beclin-1（BECN1）基因编码一个 60 kDa 的卷曲螺旋蛋白，该蛋白与原型凋亡抑制因子 Bcl-2相互作用。Beclin-1 促进 agp6/vps30 靶向缺失的自噬缺陷型酵母菌发生自噬，也促进人 MCF7 乳腺癌细胞发生自噬。Beclin-1 在 MCF7 细胞中的自噬促进活性与 MCF7 细胞增殖的抑制、体外克隆性和肿瘤发生有关，并且人们发现其可减轻帕金森′氏症和路易体病的 α-突触核蛋白模型的神经退行性病变。内源性Beclin-1蛋白在人乳腺上皮癌细胞系和组织中通常表达水平较低，但在正常乳腺上皮细胞中普遍表达水平较高。研究还表明，Beclin 1含有富含亮氨酸的核输出信号基序，从而增加了其自噬和/或肿瘤抑制功能可能需要受调节的、依赖CRM1的核质转运的可能性。
观察值〜50 kDa。在某些细胞裂解物中可能会观察到未表征的条带。

蛋白质印迹/肽抑制分析：5 µg/mL代表性批次在LNCap细胞裂解物中检测到磷酸化Beclin（Thr119）。在添加磷酸肽的情况下观察到肽抑制作用，但在添加非磷酸肽的情况下未观察到肽抑制作用。
