一般描述

辅酶A(CoA)是生命系统中必不可少的辅因子,由泛酸(维生素B5)合成。线粒体和过氧化物酶体中的CoA水平分别对应于2-5 mM和0.7 mM。胞质CoA的范围在0.05 mM至0.14 mM之间

生化/生理作用

辅酶 A(CoA、CoASH、HSCoA)是促进酶促酰基转移反应,并支持脂肪酸合成和氧化的辅酶。CoA可参与多种酶反应机制。在CoASH存在下,有机羧酸可形成酰基辅酶A硫酯,其可促进酶的识别。由异生素羧酸形成的酰基辅酶A会增加化合物的毒性,这可导致细胞代谢功能障碍。它参与Kreb′s循环中丙酮酸的氧化。CoA是代谢活动所需的成分。细菌CoA途径是抗菌药开发的靶标。这一成分可介导酰基转移和羰基活化。CoA及其硫酯水平对于细胞动态平衡至关重要。CoA还参与调节血小板聚集和血管收缩。在酶促乙酰转移反应中,它起着重要的辅助因子作用。
