General description

钙连蛋白包括一个延伸到内质网腔内的长（461个氨基酸）N端Ca²⁺-结合结构域、一个短（22个氨基酸）跨膜片段和一个酸性胞质结构域（96个氨基酸）。

钙连蛋白是钙结合 I 型整合膜蛋白，主要存在于内质网中。它在蛋白质合成和折叠期间的质量控制中起着关键作用。钙连蛋白抗体（以1:300稀释)可用于双重免疫荧光染色，显示TG2转运情况。

Biochem/physiol Actions

兔抗钙连蛋白抗体与犬、小鼠、大鼠和人的钙连蛋白（90 kDa）发生特异反应。

钙连蛋白结合新合成的糖蛋白和错误折叠的蛋白，现认为其在蛋白合成和折叠期间，在质量控制过程中起关键作用。钙连蛋白作为凝集素样伴侣蛋白，结合新合成 N-连接糖蛋白的寡糖残基。研究显示，在通过内质网（ER）转运过程中，钙连蛋白与几种细胞表面蛋白相关，这些蛋白包括主要组织相容性复合物（MHC）I类重链、T细胞受体（TCR）和B细胞膜免疫球蛋白。它还与Ca²⁺ 依赖驻留蛋白相关的其他驻留 ER 蛋白形成复合物。
