一般描述

来自人鼻病毒 14 类型的 HRV-3C 蛋白酶是一种在 8 个残基识别序列内特异性切割的蛋白酶。
蛋白水解裂解发生在 Gln 和 Gly 残基之间。
HRV-3C 蛋白酶是一种有用的工具，用于切割在载体结构域和目的蛋白之间以该序列作为融合蛋白表达的重组蛋白。
该重组版本含有 6 个组氨酸标签，可通过 IMAC 色谱法轻松去除。

应用

HRV-3C 蛋白酶已被用于去除 DsCystatin 蛋白上的 GST 标签。

一单位HRV-3C蛋白酶定义为：在0°C条件下，在反应缓冲液中每小时消化1 nM底物肽（H-Glu-Ala-Leu-Phe-Gln-pNA）所需的酶量。反应缓冲液组成为25 mM HEPES（pH 7.5）、150 mM NaCl、1 mM EDTA和1 mM DTT。
