5′-AMP-activated protein kinase subunit beta-1 (UniProt: Q9Y478; also known as AMPK subunit beta-1, AMPKb) is encoded by the PRKAB1 (also known as AMPK) gene (Gene ID: 5564) in human.The protein encoded by this gene is a regulatory subunit of the AMP-activated protein kinase (AMPK). AMPK is a heterotrimer consisting of an alpha catalytic subunit, and non-catalytic beta and gamma subunits. AMPK is an important energy-sensing enzyme that monitors cellular energy status. In response to cellular metabolic stresses, AMPK is activated, and thus phosphorylates and inactivates acetyl-CoA carboxylase (ACC) and beta-hydroxy beta-methylglutaryl-CoA reductase (HMGCR), key enzymes involved in regulating de novo biosynthesis of fatty acid and cholesterol. This subunit may be a positive regulator of AMPK activity. PRKAB1 is widely expressed in various tissues, including skeletal muscle, liver, and adipose tissue, indicating its essential role in metabolic regulation. It is primarily localized in the cytoplasm, with additional presence in the nucleoplasm and nucleus. The myristoylation and phosphorylation of this subunit have been shown to affect the enzyme activity and cellular localization of AMPK. This subunit may also serve as an adaptor molecule mediating the association of the AMPK complex. PRKAB1 is phosphorylated when associated with the catalytic subunit (PRKAA1 or PRKAA2). It is also phosphorylated by ULK1, which negatively regulates AMPK activity and suggests the existence of a regulatory feedback loop between ULK1 and AMPK. Dysregulation of PRKAB1 has been linked to several diseases, including type 2 diabetes and various cancers, where altered AMPK signaling can lead to metabolic dysfunction and tumorigenesis. Recent studies have demonstrated association of PRKAB1 mRNA expression with occurrence of coronary artery disease in type 2 diabetes, highlighting its potential as a biomarker and therapeutic target. Application
Quality Control Testing
Evaluated by Western Blotting in lysate from HeLa cells.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PRKAB1 in lysate from HeLa cells.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PRKAB1 in lysates from K-562 cells and mouse kidney tissue.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PRKAB1 in human kidney tissue sections.

Biochem/physiol Actions
Clone 2M1 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PRKAB1. It targets an epitope within 24 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
