General description
~45 kDa observed. Five isoforms between 10-45 kDa are known to exist. This antibody recognizes the P45 proenzyme (~45 kDa) and the p20 subunit (~20 kDa) of the reduced, human Caspase 1 isoforms.
Caspase-1, also known as IL-1 beta-converting enzyme (ICE), is an enzyme that cleaves the inflammatory cytokines IL-1 beta and IL-18, releasing the mature, active peptides. Caspase-1 is important for defense against pathogens and it also is involved in the induction of apoptosis. Thymocytes from caspase-1 deficient mice are resistant to Fas-mediated apoptosis, and caspase-1 is similar to the cell death gene CED-3 of C. elegans. Caspase-1 is a heterotetramer that consists of two anti-parallel heterodimers, each one formed by a 20 kDa (p20) and a 10 kDa (p10) subunit. The enzyme has a preferred cleavage site of Tyr-Val-Ala-Asp (YVAD). Caspase-1 is highly expressed in spleen and lung, and is absent from brain.

Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected Caspase 1 in human spleen tissue. Additionally, a 1:4,000 dilution from a representative lot detected Caspase 1 in human colon tissue.

This antibody recognizes the p20 subunit of Caspase 1. This antibody may cross-react with Caspase 5 due to 92% sequence homology. Additionally, this antibody recognizes the P45 proenzyme (~45 kDa) and the p20 subunit (~20 kDa) of the reduced, human Caspase 1 isoforms.
