一般描述

含蛋白水解和胶原水解酶的细胞分离溶液。可用于将常规细胞从标准组织培养塑料和粘附涂层塑料(包括SmartPlastic)上进行常规分离。 ACCUTASE 不含有哺乳动物或细菌来源产品。

由于该试剂不包含通用蛋白水解酶,因此可能无法均匀地解离所有细胞/组织类型。通常需要对细胞类型和传代次数以及孵育时间和试剂强度进行一些优化,以优化试剂的有效性。

ACCUTASE对多种细胞类型有效,包括:成纤维细胞、角质形成细胞、血管内皮细胞、肝细胞、血管平滑肌细胞、肝细胞祖细胞、原代鸡胚神经元细胞、骨髓干细胞、贴壁CHO和BHK细胞、巨噬细胞、293细胞、L929细胞、永生小鼠睾丸生殖细胞、3T3、Vero、COS、HeLa、NT2、MG63、M24和A375转移性黑色素瘤、神经胶质瘤U251和D54、HT1080纤维肉瘤细胞以及Sf9昆虫细胞。

每批次的ACCUTASE都已经过无菌性(通过USP膜过滤方法)、酶活性(通过合成发色四肽测试)和细胞与组织培养塑料的分离性能测试。

应用

细胞分离

1.将ACCUTASE®在室温下解冻。

2.用无菌PBS洗涤孔板、烧瓶或珠子。

3.通过无菌程序添加ACCUTASE至培养皿或烧瓶中(每75平方厘米表面积10 ml)。

4.经培养物放回37°C培养箱中,以分离细胞(5-10分钟)。

5.按常规操作进行细胞计数和传代。无需额外的洗涤或酶抑制剂



每种细胞/组织类型的最佳孵育时间和试剂强度应有最终用户进行确定。

外形

100 ml,即用型,冷冻无菌液体。 1X ACCUTASE酶,溶于含0.5 mM EDTA·4Na和3 mg/L苯酚红的Dulbecco′s PBS(0.2 g/L KCl、0.2 g/L KH2PO4、8 g/L NaCl和1.15 g/L Na2HPO4)中。

储存及稳定性

保存于-20°C可保持稳定。请参阅批次到期日期。建议收货时保存于-20°C。解冻后,ACCUTASE可以在4°C下最多保存2个月。请勿在室温下存放
