一般描述
DNase I是一种可降解DNA的双链特异性内切核酸酶。牛胰腺脱氧核糖核酸酶I（DNase I）是DNA小沟相互作用的核酸酶，特异性相对较低。该蛋白质由两个中心β折叠构成，而每个折叠由六个β-链所组成。该结构被大量的环以及α-螺旋区域包围。该酶与核酸外切酶III具有结构相似性。DNase I是研究得最透彻的哺乳动物来源的内切酶之一。
生化/生理作用
在组织解离过程中，部分细胞被裂解并导致DNA的释放。单分子DNA可能会导致细胞结团。向解离缓冲液中加入DNase I会导致该细胞外DNA的降解，因而避免了因意外结团造成的细胞损失。
单位定义
一个单位表示在测定条件下导致吸光度以0.001每分钟的速率而增加的酶活力。
制备说明
激活剂：二价金属离子 (Ca2+, Mg2+)
工作浓度：0.01 至 1 mg/ml
注意：必须针对每种细胞类型单独确定工作浓度。为实现最佳活性，酶须添加 5 mM Mg2+。

重悬
用无菌的双蒸水对冻干酶进行重悬（10 mg/ml）；用PBS（磷酸盐缓冲液）、HBSS（Hank′s 平衡盐溶液）或培养基进行进一步的稀释。
