General description

硫酸乙酰肝素蛋白多糖（HSPG）是一种糖蛋白，是基底膜不可或缺的一部分。它是由通过共价键结合硫酸肝素链的核心蛋白组成。HSPG有三种：膜HSPG、细胞外基质分泌HSPG和分泌泡蛋白多糖。

Application

硫酸乙酰肝素蛋白多糖已用于:

  • 在一项研究中加入磷酸缓冲生理盐水(PBS) ，并添加至含柠檬酸盐的新鲜血样中以获得与创伤等效（trauma-equivalent ）的浓度
  • 固相结合测定

Biochem/physiol Actions

硫酸乙酰肝素蛋白多糖（HSPG）在多个生物过程中发挥多重功能。它们有助于形成基底膜结构并为细胞迁移提供基质。在分泌泡中，如丝甘蛋白聚糖，HSPG在颗粒状内容物的包裹、蛋白酶活性的维持、分泌后生物活性的调节方面起一定作用。HSPG可以结合和保护细胞活素、生长因子和趋化因子，有助于形成对于细胞特异性分化和白细胞募集必不可少的梯度。它们可作为蛋白酶受体和蛋白酶抑制剂，调节细胞-细胞外基质附着、细胞-细胞相互作用和细胞运动，可与生长因子受体协同作用。HSPG也可作为配体清除中的胞吞受体，并参与脂蛋白代谢和形态发生素的梯度形成。也是基质溶解因子和其他基质金属蛋白酶的对接分子。硫酸乙酰肝素可促进培养细胞附着、诱导碱性FGF与缺乏硫酸肝素和可溶性FGF受体的细胞高亲和力结合，工作浓度为10-100 ng/mL。

Preparation Note

在含有50 mM Tris HCl（pH 7.4）、150 mM NaCl、1 mM EDTA和0.1 mM PMSF的溶液中，从Engelbreth-Holm-Swarm小鼠肉瘤的基底膜中分离得到该产品。
