General description
本品为 0.2 μm 过滤水溶液，含 1 mg/mL 来自大肠杆菌血清型 O55:B5 的脂多糖 (LPS)。来源菌株为 CDC 1644-70。这种 LPS 血清型已被用于感染性休克和心肌炎的研究。
Application
脂多糖（LPS）是革兰氏阴性细菌细胞壁的特征组分。脂多糖及其脂质 A 部分通过 Toll 样受体 4（TLR4）刺激先天免疫系统的细胞，Toll 样受体 4是 Toll 样受体蛋白家族的成员，其识别常见的病原体相关分子模式（PAMP）。
Biochem/physiol Actions
脂多糖位于膜的外层，在非包膜菌株中，暴露在细胞表面。它们有助于外膜的完整性，保护细胞免受胆盐和亲脂性抗生素的作用。
Preparation Note
用于制备溶液的 LPS 经过苯酚提取和凝胶过滤层析纯化。通过苯酚提取制备的脂多糖含有高达 60% 的 RNA 和少于 1% 的蛋白质。随后通过凝胶过滤色谱法进行纯化，可去除苯酚提取的 LPS 中存在的大部分蛋白质，但留下的产物仍含有 10-20% 的核酸。采用离子交换色谱法进一步纯化，得到含 < 1% 蛋白质和 < 1%RNA 的 LPS 产品。
