General description

邻苯二胺二盐酸盐是双亲核试剂，其与α, β-不饱和羰基化合物和醛的缩合产生苯并咪唑。
邻苯二胺二盐酸盐（OPD）是一种显色底物，因为利用辣根过氧化物酶缀合物，因此可用于酶联免疫吸附试验（ELISA）程序。底物产生可溶的最终产物，其颜色为橙棕色，并且可以在450 m下通过分光光度计读取。可以用3N HCl或3M H2SO4终止OPD反应，并在492 nm处读取。

Application

邻苯二胺二盐酸盐已被用作辣根过氧化物酶的底物，用于检测：

Preparation Note

将 1 片片剂溶于 0.05 M 磷酸盐-柠檬酸盐缓冲液 (pH 5.0) 中，得到所需浓度（通常 OPD 浓度为 使用 0.4 mg/mL）。在使用前立即每 100ml 底物缓冲溶液中加入 40μl 新鲜的 30% 过氧化氢。片剂分别包装在箔包中。
