General description

Application
凝血酶用于位点融合蛋白的位点特异性裂解，重组融合蛋白含有可进入的凝血酶识别位点，用于去除亲和标签。在一项研究中，凝血酶被用于评估用于生物物理学和结构表征的腺苷受体肽生物合成的表达和纯化系统。
Biochem/physiol Actions
丝氨酸蛋白酶，选择性断开纤维蛋白原中的精氨酸-甘氨酸键，形成纤维蛋白以及纤维蛋白肽 A 和 B。
Preparation Note
当使用 1mL 水复原时，小瓶将在 0.15M 氯化钠和 0.05M 柠檬酸钠中以及 pH 6.5 下达到规定的活性。
Analysis Note
基于 Biggs 改性法的 NIH 分析过程使用 0.2mL 稀释血浆（由血浆与等量生理盐水混合制得）作为底物，以及 0.1mL 凝血酶样品（在含 1% 白蛋白缓冲液中保持稳定）。只使用 15-25 秒之间的凝血时间分析凝血酶浓度。
活性是通过与一种K批次的NIH凝血酶参考标准品进行直接比较而得到并以NIH单位进行表达的。
